Molecular Interactions between CD21/35 & the Actin Cytoskeleton in Follicular Dendritic Cells are Regulated by Cell-ECM Traction Forces

Follicular dendritic cells (FDCs) are stromal that reside in follicles of secondary and tertiary lymphoid organs germinal centres. They provide structure to the tissue micro-environment promote proliferation, activation, affinity maturation B cells. FDCs express high levels complement receptor 1/2 (CD21/35) on their surface retain immune complexes for presentation It has been suggested FDC actin cytoskeleton influences sensitivity specificity cell responses, but underlying mechanisms not clear. In this project we investigating interplay between CD21/35 clustering, diffusion, cortical dynamics mouse primary how is influenced by traction forces exerted onto extracellular matrix (ECM). Using a combination drug inhibitors activators, show filamentous regulates spatial organisation mobility membrane. Moreover, culturing polyacrylamide hydrogels different stiffness modulate FDC-ECM forces, respond changes substrate altering size structures, which impacts its association with actin.
